Ethical Clearance and Trial Overview

Patronus Biotech announced that the Bellberry Limited Research Ethics Committee granted ethical approval on 19 October 2026 for its first‑in‑human Phase I clinical study (registered as NCT07831226). The study will evaluate three novel multi‑stage virus‑like‑particle (VLP) malaria vaccine candidates—LYB014, LYB017 and LYB027—targeting key stages of the Plasmodium falciparum and Plasmodium vivax life cycles. The trial is designed as a randomised, double‑blinded, controlled, dose‑escalation study and will enrol healthy adult volunteers at Nucleus Network Pty Ltd., located in Melbourne, Australia, with the first participant expected to be enrolled in December 2026.

Vaccine Candidates and Adjuvant

LYB014 is engineered to address three critical stages of the P. falciparum parasite, LYB017 targets the analogous three stages of P. vivax, and LYB027 is a combined candidate that simultaneously targets the three stages of both species. All three vaccines will be administered with Patronus Biotech’s proprietary A02 adjuvant, a novel liposomal formulation that contains fully synthetic active ingredients, intended to enhance immunogenicity across the multiple parasite stages.

Immunogenicity Analysis Partnership

The immunogenicity read‑outs from the trial will be performed in collaboration with the Draper Lab at the Department of Paediatrics, University of Oxford. Professor Simon Draper, Professor of Vaccinology and Translational Medicine, expressed enthusiasm for the partnership, noting the ongoing global need for broader malaria vaccine approaches.

Rationale and Context

Malaria continues to pose a major global health challenge, with an estimated 282 million cases and 610 000 deaths reported in 2024, primarily caused by P. falciparum and P. vivax. Existing WHO‑approved vaccines—RTS,S/AS01 and R21/Matrix‑M—target only the sporozoite (first) stage of P. falciparum and have shown partial efficacy in paediatric field trials. Patronus Biotech’s multi‑stage strategy seeks to determine whether adding protection against liver‑stage, blood‑stage, and sexual‑stage parasites, and extending coverage to both species, can provide layered, complementary immunity that could contribute to malaria elimination efforts.

Executive Statements

Dr. Jin, Chief Executive Officer of Patronus Biotech, stated that the company’s goal is to develop a more robust and affordable shield against malaria by targeting the parasite’s complex biology. He highlighted that the combination of the three VLP candidates with the A02 adjuvant aims not only to prevent infection but also to reduce disease severity and transmission.
