FDA Accelerated Approval of GENGLYCOS

Ultragenyx Pharmaceutical Inc. (NASDAQ:RARE) received accelerated approval from the U.S. Food and Drug Administration for GENGLYCOS, also known as DTX401, marking the company’s first gene‑therapy authorization and its fifth FDA approval overall. The therapy is approved for adult and pediatric patients aged eight years and older diagnosed with glycogen storage disease type Ia (GSDIa), an ultra‑rare metabolic disorder that affects an estimated 1,500‑2,500 individuals in the United States. In addition to the approval, Ultragenyx was granted a Priority Review Voucher.

Clinical Trial Evidence

The FDA’s decision was based on positive results from the 48‑week Phase 3 GlucoGene study, a randomized, double‑blind, placebo‑controlled trial that enrolled 46 participants aged eight years and older. The study demonstrated a reduction in the amount of raw cornstarch required by patients receiving GENGLYCOS compared with those on placebo, indicating improved glucose management for this population.

Post‑Approval Obligations

As a condition of the accelerated approval, Ultragenyx agreed to provide two years of safety and efficacy data from an open‑label commercial treatment cohort comprising 50 patients receiving GENGLYCOS and a control cohort of 20 patients who cannot be treated due to anti‑AAV8 antibodies. These data will be collected through the company’s existing GSDIa Disease Monitoring Program.

Commercial Availability

GENGLYCOS will be distributed through a national network of Qualified Treatment Centers that possess specialized expertise in gene‑therapy administration. The therapy is manufactured at Ultragenyx’s Gene Therapy Manufacturing Facility located in Bedford, Massachusetts.
